BAOJ Microbiology Molecular Affirmation of Aspergillus luchuensis a Niger Group Species as a Non- Ochratoxigenic Fungus

چکیده
Amongst the various species of Aspergillus section Nigri; Aspergillus luchuensis, associated with food and beverage fermentation in East Asia, is evaluated by multiple researchers as nonochratoxigenic based on the extrolite analysis. Beyond extrolite analysis, in the present study, A. luchuensis strain was analyzed by a rapid Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) technique. The Internal Transcribed Spacer (ITS), Intergenic Spacer (IGS) and β-tubulin gene region were PCR amplified from A. luchuensis. The digestion of amplicons ITS and β-tub with Rsa I resulted in uncut and two bands (420 and 130 bp) respectively, while IGS with Hinf I produced three bands (250, 80 and 60 bp). The restriction profile obtained, resembled the profile of strains which lack the ability to produce ochratoxin A (OTA). A clear correlation between the extrolite analysis and PCRRFLP technique is established, confirming the usability of PCRRFLP method for the rapid affirmation of non-ochratoxigenic property of A. luchuensis.

Aspergillus Luchuensis, an Industrially Important Black Aspergillus in East Asia
Aspergilli known as black- and white-koji molds which are used for awamori, shochu, makgeolli and other food and beverage fermentations, are reported in the literature as A. luchuensis, A. awamori, A. kawachii, or A. acidus. In order to elucidate the taxonomic position of these species, available ex-type cultures were compared based on morphology and molecular characters. A. luchuensis, A. kawa...
متن کاملOchratoxigenic fungi associated with green coffee beans (Coffea arabica L.) in conventional and organic cultivation in Brazil
The genera Aspergillus comprises species that produce mycotoxins such as aflatoxins, ochratoxins and patulin. These are cosmopolitan species, natural contaminants of agricultural products. In coffee grains, the most important Aspergillus species in terms of the risk of presenting mycotoxins belong to the genera Aspergillus Section Circumdati and Section Nigri. The purpose of this study was to a...
متن کاملAnalysis of the MAT1-1 and MAT1-2 Gene Ratio in Black Koji Molds Isolated from Meju
Aspergillus luchuensis is known as an industrially important fungal species used for making fermented foods such as awamori and shochu in Japan, makgeolli and Meju in Korea, and Pu-erh tea in China. Nonetheless, this species has not yet been widely studied regarding mating-type genes. In this study, we examined the MAT1-1 and MAT1-2 gene ratio in black koji molds (A. luchuensis, Aspergillus nig...
متن کاملBlack Aspergillus species as ochratoxin A producers in Portuguese wine grapes.
To evaluate the incidence of fungi producing ochratoxin A (OA) in Portuguese wine grapes, a survey was conducted in 11 vineyards, from four winemaking regions each with distinct climatic conditions. From setting to the harvesting period, a total of 1,650 berries were sampled by plating methods. Out of 370 aspergilli and 301 Penicillium strains isolated, 14% of the aspergilli were OA-producing s...

A total of 150 cereal samples from three regions of Southern India i.e. Karnataka, Andhra Pradesh and Tamilnadu were collected during winter 2010 – 2012 to determine the ochratoxigenic fungal incidences. A total of 150 fungal isolates comprising, Aspergillus ochraceus, A. niger and Penicillium verrucosum were recovered and tested for ochratoxin A (OTA) production. Chemotype determination of fun...
